No, territories will act on a lead (to assign an owner on a round-robin basis), whenever the condition of the territory is satisfied and when the territory and owner fields are empty. If there is an owner assigned and if the territory is empty, then the territory value will be updated on the lead but the round-robin owner assignment will not work (since the owner has already been assigned)
